Lab Presentation

Our research focuses on the structure-function relationships of proteins and macromolecular complexes directly implicated in pathological processes. In particular, virus particles, viral proteins and protein-complexes involved in RNA replication as well as other large macromolecular assemblies. The information obtained would be instrumental not only for improving the knowledge about the functioning of these biological systems but also in the development of novel therapeutic strategies. We use a number of molecular and structural biology techniques. In particular, X-ray crystallography and combinations of X-ray crystallography and Cryo-electron microscopy.

The group has strong expertise in the determination of macromolecular structures by X-ray diffraction techniques, combined with other macromolecular analysis tools like EM. Our approaches usually start with the cloning of relevant genes and the expression and purification of the encoded proteins.

Proteins, nucleic acids and their complexes are then crystallized and analyzed by X-ray diffraction. The final outcome is a detailed three dimensional view of the molecular structures at atomic resolution. Our research interests cover three main topics:

Structural and functional characterization of different processes in the biological cycle of RNA viruses, including the structure of viral capsids, virus-receptor interactions, viral uncoating and viral replication.

Our major scientific contributions comprise:

a. The structural characterization of the first steps of human rhinovirus infection, including the first high resolution X-ray structures of a human minor group rhinovirus bound to its cellular receptor protein and the first high resolution X-ray structures of the virus trapped in the process of RNA uncoating which allowed to identify the structural alterations that take place in the capsid architecture during infection

The work included: The structures of a protein primer-polymerase complexes involved in the initiation of viral genome replication and the structure of different RNA elongation complexes, addressing important questions related with regulation of viral replication, fidelity and virus evolution .

Structural analysis of the vault ribonucleoprotein complex, the largest particle found in eukaryote.

Regarding the hypothetical functions of this particle, it has proposed to be implicated in a diversity of cellular processes, including multidrug resistance, transport mechanisms and cell signaling. However, their exact role in any of these pathways remains elusive. Major contributions to the topic are the characterization of the structure and dynamics of the Vault shell, deciphering the regulation of vault opening .Virus and Large Biological Complexes, SBU

 

Characterization of complexes involved the regulation of synaptic vesicle fusion.

We are combining multi-disciplinary integrated approaches to elucidate how Rabphilin 3A, a protein involved in vesicle fusion at the presynaptic level, coordinates its interactions with Ca2+, PIP2, SNAP25, Rab3/27 and a-synuclein to control key events at the presynaptic neuron. A recent contribution comprises the high resolution X-ray structure of the C2A domain of rabphilin3A in complex with PIP2, defining a new motif in C2 domains that determines their ability to interact with different phosphoinositides and its involvement in the reorganization of the plasma membraneVirus and Large Biological Complexes, SBU

Núria Verdaguer is the Scientific Director of the of the Maria de Maeztu Structural Biology Unit at the Institut de Biologia Molecular de Barcelona (IBMB-CSIC).

She obtained her degree in Biological Sciences from the University of Barcelona (UB) in 1986 and her Ph.D. in 1991 from the Polytechnic University of Catalonia (UPC). In 2002 she started a structural virology research group at the IBMB-CSIC and promoted to full Professor in 2009.

Núria Verdaguer was elected member of the European Molecular Biology Organization (EMBO) in 2008. She has been director of the Department of Structural Biology from October 2010 to September 2012 and Deputy Director of the IBMB from July 2014 to June 2016.
